Formal discussion of: somatic mutations and carcinogenesis.

چکیده

A great deal of evidence is now available indicating that mutations play a dominant role in the aging process. Because carcinogenesis is one facet of the aging process, mu tations must play a major role here also. However, carcinogenesis certainly does not consist of the simple induction of a mutation in one cell which confers the property of uncontrolled growth on that cell and its progeny. There must be some general condi tions, as well as a specific event, before carcinogenesis can take place. It is postulated that mutations in one form or another are responsible for both conditions.

Tumorogensis : The Dual Role of Telomerase

Carcinogenesis is a multistep process characterized by the gradual accumulation of genetic changes that ultimately lead to cancer. These genetic mutations can impart limitless replicative potential to the cancer cells making them immortal. Telomeres are repeat nucleotide sequence TTAGGG that are present at the end of chromosomes. Its functions are to protect the chromosomal ends and to ensur...

The chronological sequence of somatic mutations in early gastric carcinogenesis inferred from multiregion sequencing of gastric adenomas

Mutation profiles and intratumoral heterogeneity are not well understood for benign gastric adenomas, some of which progress into malignant gastric adenocarcinomas. In this study, we performed whole-exome sequencing of three microsatellite stable (MSS) and two microsatellite instability-high (MSI-H) gastric adenomas with three regional tumor biopsies per case. We observed that the mutation abun...

Somatic Cell Mutation Assay at the Glycophorin-a Locus in Human Erythrocytes for Biodosimetry

For risk assessment, it is necessary to evaluate the dose of human exposure to radiation. Ionizing radiation being an environmental genotoxic factor induces dosedependent somatic mutations. Recent studies support the somatic-mutation theory of carcinogenesis. Mutations at the Glycophorin–A (GPA) loci have been studied for establishment of somatic mutation assays. GPA assay provides a lifetime b...

Stochastic modeling indicates that aging and somatic evolution in the hematopoietic system are driven by non-cell-autonomous processes

Age-dependent tissue decline and increased cancer incidence are widely accepted to be rate-limited by the accumulation of somatic mutations over time. Current models of carcinogenesis are dominated by the assumption that oncogenic mutations have defined advantageous fitness effects on recipient stem and progenitor cells, promoting and rate-limiting somatic evolution. However, this assumption is...

Somatic Host Cell Alterations in HPV Carcinogenesis

High-risk human papilloma virus (HPV) infections cause cancers in different organ sites, most commonly cervical and head and neck cancers. While carcinogenesis is initiated by two viral oncoproteins, E6 and E7, increasing evidence shows the importance of specific somatic events in host cells for malignant transformation. HPV-driven cancers share characteristic somatic changes, including apolipo...
